Gold Labeling for Cryoelectron Microscopy and Electron Tomography

چکیده

Electron Tomography Cryotechniques are a hot topic right now, and the introduction of new preparation methods has allowed electron microscopists insights into organelles and other biological structures that are not possible with traditional embedding methods. For example, cryoelectron tomography of frozen-hydrated specimens reveals that the traditional picture of spatial connectivity in mitochondria is inaccurate, and FRIL (Freeze-Fracture Immunogold Labeling) has been used to probe the ultrastructure of gap junctions. Instruments for reliable high-pressure freezing have enabled correlative microscopy with improved preservation, and optimized methods for freeze substitution are also enabling new discoveries.
